II. Working Principle: Goalkeeper Mechanism

DRAQ7 is a charged anthraquinone derivative that cannot penetrate the intact plasma membrane of live cells – this "gate" separates live cells from the outside, and DRAQ7 is kept out. When cells undergo apoptosis, necrosis, or membrane damage, the "gate" is breached, allowing DRAQ7 to enter the nucleus and intercalate into the double helix structure of double-stranded DNA. Upon intercalation, the fluorescence is significantly enhanced, emitting far-red light with an emission wavelength of 694-697 nm.

A simple three-step logic chain: Structure (membrane-impermeable anthraquinone) → Event (membrane damage allows entry) → Signal (fluorescence turns on upon DNA intercalation). This mechanism ensures that only dead or membrane-damaged cells are labeled.
