II. Working Principle

This class of substrates is constructed as arylamide peptide substrates by forming an amide bond between the C-terminus of the X-Arg peptide residue and the 4-methoxy-β-naphthylamine (4MβNA) fluorescent reporter group. In the intact state, the fluorescence of 4MβNA is quenched, thus no fluorescence signal is released.

Taking coagulase detection as an example, signal release occurs in three steps:

1. Recognition and Cleavage: Coagulase specifically recognizes the arylamide peptide bond in the substrate, catalyzes the hydrolysis of the peptide bond, and releases 4MβNA from the peptide chain.

2. Fluorescence Activation: Free 4MβNA emits strong fluorescence (at ~410 nm) upon excitation at ~340 nm. The higher the enzyme activity, the more 4MβNA is released per unit time, and the faster the fluorescence intensity increases.

3. Quantitative Interpretation: By continuously monitoring the rate of change in fluorescence intensity using a fluorescence microplate reader or fluorescence spectrophotometer, the coagulase activity unit (U/mL) can be accurately calculated, achieving a seamless transition from "microsample → minute-level readout → objective data".



III. Application Scenarios

• Coagulation Research: Used for accurate detection of coagulase activity, helping to reveal blood coagulation mechanisms and related diseases;

• Vaginal Infection Typing: Bacterial coagulase positivity can serve as an important indicator suggesting Staphylococcus aureus infection in vaginal infection typing;

• Dipeptidyl Peptidase I (DPP-I) Analysis: Also detects dipeptidyl peptidase I, applicable for studying lysosomal function in leukemia cell lines;
